Where to startMolecular biology methods and biophysical methods together carry more than half the unit, and most of those questions are experiments to interpret: primer design, restriction maps, blots and gels, footprinting, CD spectra and mass spectra. The statistics questions are among the most predictable in the paper. Learn which test fits which design (parametric or not, paired or not) and what a confidence interval means. Microscopy questions keep coming back to resolution (0.61λ/NA) and to which super-resolution method needs which probe.

What type of electromagnetic radiation is used in biomolecular NMR spectroscopy?

  1. 1Radio waves
  2. 2Beta emissions
  3. 3X-ray waves
  4. 4Microwaves
Show answer and solution
Answer: (1) Radio waves

NMR detects transitions between nuclear spin states in a strong magnetic field. The energy gap is tiny, so the resonance frequencies are in the radio-frequency range, hundreds of MHz (for example 600 MHz for 1H on a 14.1 T magnet). Microwaves are used in ESR/EPR, which detects electron spins.

Which one of the following combinations of excitation light and objectives gives the best lateral resolution in a fluorescence microscope?

  1. 1480nm, 63X, 1.4NA
  2. 2650nm, 63X, 1.4NA
  3. 3550nm, 100X, 1NA
  4. 4480nm, 100X, 1.1NA
Show answer and solution
Answer: (1) 480nm, 63X, 1.4NA

Lateral resolution follows the Abbe/Rayleigh limit, d = 0.61λ/NA. Smaller is better, so you want the shortest wavelength and the highest numerical aperture. Magnification does not set resolution. 480 nm with NA 1.4 gives d ≈ 209 nm, better than the other three combinations.

Q70Microscopic TechniquesJun 2023 S1 · Part B · 2 marks, −0.5

Which one of the following factors will NOT have any impact on the resolving power of a bright field microscope?

  1. 1Color of light
  2. 2Intensity of light
  3. 3Angle of admittance of light in the objective lens
  4. 4Medium between the objective lens and specimen
Show answer and solution

Answer: (2) Intensity of light

Resolving power depends on the wavelength (the colour of light), the half-angle of light the objective accepts, and the refractive index of the medium. The last two together make up the numerical aperture. The intensity of light changes brightness and contrast, but not the smallest distance that can be resolved.

Q69Microscopic TechniquesFeb 2022 S1 · Part B · 2 marks, −0.5

Fluorescence microscope that requires photoactivatable probes to obtain super-resolution is

  1. 1Structured illumination microscope (SIM)
  2. 2dSTORM – stochastic optical reconstruction microscopy
  3. 3Stimulated emission depletion microscopy (STED)
  4. 4Laser scanning confocal microscope
Show answer and solution

Answer: (2) dSTORM – stochastic optical reconstruction microscopy

dSTORM, like PALM, depends on single-molecule localisation. Photoswitchable or photoactivatable fluorophores are made to blink so that only a few, well-separated molecules are on in each frame. Their centres are found to nanometre precision and built up into an image. STED and SIM reach super-resolution by shaping the illumination and work with ordinary dyes.

Q68Microscopic TechniquesDec 2023 S2 · Part B · 2 marks, −0.5

Which microscope is typically used to detect a single fluorescent molecule?

  1. 1DIC microscope
  2. 2Epifluorescence microscope
  3. 3TIRF microscope
  4. 4Phase contrast microscope
Show answer and solution

Answer: (3) TIRF microscope

In TIRF microscopy, light totally internally reflected at the coverslip produces an evanescent field that reaches only about 100 nm into the sample. Out-of-focus fluorescence from the rest of the cell is never excited, so the background is low enough to see single fluorophores near the membrane.

A pulse oximeter measures the ratio between oxyhemoglobin and hemoglobin in the pulsating condition on the finger. Which combination of wavelengths can obtain a differential absorption to measure blood oxygen saturation?

  1. 1Ultraviolet and Infrared
  2. 2Visible and Infrared
  3. 3Far-ultraviolet and visible
  4. 4Ultraviolet and far-ultraviolet
Show answer and solution

Answer: (2) Visible and Infrared

Oxyhaemoglobin and deoxyhaemoglobin absorb differently in visible red light (about 660 nm), where deoxyhaemoglobin absorbs more, and in near-infrared (about 940 nm), where oxyhaemoglobin absorbs more. The oximeter compares the pulsatile part of the absorbance at the two wavelengths to work out saturation.

Q89Biophysical MethodsDec 2024 S2 · Part C · 4 marks, −1

A molecule absorbs light at ‘X’ nm wavelength and emits light as fluorescence at ‘Y’ nm wavelength. Typically, there is a shift in the wavelength (Y>X). E is the energy transferred to the solvent during reorganization of the excited state, ‘h‘ is the Planck’s constant, and ‘c’ is the speed of light.

E is equal to:

  1. 1h(Y−X)
  2. 2hc(Y−X) / XY
  3. 3c(Y−X) / h
  4. 4c / (Y−X)
Show answer and solution

Answer: (2) hc(Y−X) / XY

Photon energy is E = hc/λ. Light is absorbed at X and emitted at the longer wavelength Y, so the energy lost in between (the Stokes shift, passed to the solvent) is hc/X − hc/Y = hc(Y − X)/XY.

Which one of the following techniques can identify acetylation on a lysine residue of a protein?

  1. 1Mass spectrometry
  2. 2SDS-PAGE
  3. 3Light scattering
  4. 4CD spectroscopy
Show answer and solution

Answer: (1) Mass spectrometry

Acetylation adds exactly 42.011 Da to a lysine. Mass spectrometry, usually tandem MS of a digest, detects that mass shift and finds the modified residue from the fragment ions. (It is told apart from trimethylation, +42.047 Da, by high mass accuracy.) SDS-PAGE, light scattering and CD cannot see a 42 Da change at a single site.

In rate-zonal centrifugation, a mixture is spun just long enough to separate molecules that differ in mass but may be similar in shape and density. Which one of the following options represents the component that is typically used to create a gradient in this technique?

  1. 1phospholipids
  2. 2sucrose
  3. 3KCl
  4. 4starch
Show answer and solution

Answer: (2) sucrose

Rate-zonal centrifugation separates particles by how fast they sediment through a shallow, pre-formed density gradient, usually sucrose (for example 5–20%). The gradient stabilises the bands against convection. The run is stopped before anything reaches equilibrium. Isopycnic separations use CsCl instead.

The units of molar extinction coefficient are

  1. 1L mol−1cm−1
  2. 2cm−1mg ml−1
  3. 3mol−1mm
  4. 4mol cm ml−1
Show answer and solution

Answer: (1) L mol−1cm−1

From the Beer–Lambert law, A = εcl. Absorbance has no units, so ε = A/(c × l), with c in mol L−1 and l in cm. That gives L mol−1 cm−1 (M−1 cm−1).

Which one of the following is the most sensitive immunoassay?

  1. 1Immuno-electrophoresis
  2. 2Rocket electrophoresis
  3. 3Immunofluorescence
  4. 4Passive agglutination
Show answer and solution

Answer: (4) Passive agglutination

In the classical comparison of immunoassay sensitivities (Kuby), passive agglutination, with antigen-coated red cells or latex beads, detects antibody at about 0.006–0.06 µg/mL. Immunofluorescence detects about 1 µg/mL, rocket electrophoresis about 2, and immunoelectrophoresis needs 20–200. Among these options, passive agglutination is the most sensitive.

Which of the following molecular marker techniques uses a single primer in a PCR reaction for identifying polymorphisms between genotypes?

  1. 1SSR (Simple sequence repeats)
  2. 2AFLP (Amplified fragment length polymorphism)
  3. 3RAPD (Randomly amplified polymorphic DNA)
  4. 4SCAR (Sequence characterized amplified regions)
Show answer and solution

Answer: (3) RAPD (Randomly amplified polymorphic DNA)

RAPD amplifies genomic DNA with a single short (about 10-mer) arbitrary primer. Wherever two copies of it bind close together on opposite strands, a band forms, so polymorphisms show up as bands that are present or absent. SSR and SCAR use specific primer pairs, and AFLP uses adaptor-specific primers after restriction digestion and ligation.

Q43Statistical MethodsJun 2024 S2 · Part B · 2 marks, −0.5

Which one of the following is an example of parametric statistical test?

  1. 1Kruskal Wallis test
  2. 2Fisher’s exact test
  3. 3Unpaired t-test
  4. 4Wilcoxon signed rank test
Show answer and solution

Answer: (3) Unpaired t-test

A parametric test assumes a distribution, usually normal, and compares its parameters, such as means. The unpaired t-test is parametric. Kruskal–Wallis and the Wilcoxon signed-rank test work on ranks, and Fisher’s exact test works on counts in a contingency table, so all three are non-parametric.

Which one of the following statements is correct with respect to 95% confidence interval of the estimated mean from a set of observations?

  1. 1They are limits between which, in the long run, 95% of observations fall.
  2. 2They are a way of measuring the precision of the estimate of the mean.
  3. 3They are limits within which, the sample mean falls with probability 0.95.
  4. 4They are a way of measuring the variability of a set of observations.
Show answer and solution

Answer: (2) They are a way of measuring the precision of the estimate of the mean.

A 95% confidence interval measures the precision of the estimated mean. If sampling were repeated many times, 95% of intervals built this way would contain the true population mean. It does not describe where 95% of individual observations fall (that is spread, measured by the SD). It also says nothing about where the sample mean falls, since the interval is centred on it.

Find the linear regression equation for the following data pairs (x, y) given in the above table

  1. 1y = 4x + 0
  2. 2y = 3x + 2
  3. 3y = 6x + 2
  4. 4y = 0.33 + 2
Show answer and solution

Answer: (2) y = 3x + 2

Every step of 2 in x raises y by 6, so the slope is 6/2 = 3. At x = 0, y = 2, which is the intercept. All the points lie exactly on y = 3x + 2 (check: x = 14 gives 44).

Q57Statistical MethodsJun 2025 · Part B · 2 marks, −0.5

Which one of the following datasets (expressed as mean ± SD) is most likely to follow a Gaussian (normal) distribution, considering that the data cannot have any negative values?

  1. 1296 ± 190 picomolar concentration of insulin in human blood samples
  2. 258 ± 45 number of a particular species of bacteria in the rhizosphere
  3. 3160 ± 20 mg/dL of blood cholesterol concentration in human athletes
  4. 44 ± 8 number of insects caught in a trap from an agricultural field
Show answer and solution

Answer: (3) 160 ± 20 mg/dL of blood cholesterol concentration in human athletes

For a variable that cannot go negative, a normal distribution is plausible only if the mean lies several SDs above zero. For 160 ± 20 the mean is 8 SD from zero, so the data can be symmetric. In the other three the SD is large relative to the mean (190/296, 45/58, 8/4), so the data must be skewed to the right.
